Loss of control of epithelial basal extrusion leads to tumor resistance in Drosophila

Basal extrusion allows an epithelial cell to leave its compartment, making it a necessary step in the local invasion of all epithelial cancers. Here, we determined which effect sex steroid deprivation plays on basal extrusion, and the resulting consequences on tumorigenesis. For this, we blocked ecdysone signaling in a model of epithelial tumorigenesis in the drosophila accessory gland, in a tumor-cell specific manner. As for human treatment, this induces apoptosis of many preexistent tumor cells. It also slightly decreases primary tumor formation. However, it leads to the appearance of a new population of highly proliferative tumor cells following the loss of control of a newly discovered step of basal extrusion. Our findings suggest that steroid deprivation actively induces resistance by diverting highly controlled basal extrusion.
